E-mail: [email protected] A commonly used genome fragmentation method in next generation sequencing, restriction endonuclease (RE) digestion, may severely compromise genomic mapping resolution and prevent the functional annotation of certain chromosomal regions unless REs are applied in correct combinations to sample all genomic regions with an equal probability.
